Combined high-throughput library screening and next generation RNA sequencing uncover microRNAs controlling human cardiac fibroblast biology

BackgroundMyocardial fibrosis is a hallmark of the failing heart, contributing to most common causes deaths worldwide. Several microRNAs (miRNAs, miRs) controlling cardiac were identified in recent years; however, more global approach identify miRNAs involved missing.Methods and resultsFunctional miRNA mimic library screens applied human fibroblasts (HCFs) annotated inducing proliferation. In parallel, deep sequencing was performed after subjecting HCFs proliferating resting stimuli, additionally enabling discovery novel miRNAs. In-depth vitro analysis confirmed pro-fibrotic nature selected, highly conserved miR-20a-5p miR-132-3p. To determine downstream cellular pathways their role fibrotic response, targets candidates modulated by synthetic siRNA. We here provide evidence that repression autophagy detoxification reactive oxygen species miR-132-3p explain some on mechanistic level.ConclusionWe both as crucial regulators an model fibroblast biology.
